La créatine, [NH2-C(NH)-NCH2(COOH)-CH3], est un acide aminé naturel, présent principalement dans les fibres musculaires et le cerveau. Elle joue un rôle dans l'apport d'énergie aux cellules musculaires et dans la contraction musculaire. Le mot vient du grec kreas (viande).
Normalement la moitié de la créatine du corps humain provient de la nourriture (principalement de la viande et du poisson), alors que l'autre moitié est synthétisée à partir de certains acides aminés (glycine, arginine) dans le foie, le pancréas et les reins. 95% d'entre elle est stockée dans les muscles du squelette, le reste dans le cerveau, le cœur et les testicules.
Dans les muscles, une fraction de la créatine totale se lie au phosphate. La réaction est catalysée par la créatine kinase, et résulte en phosphocréatine (PCr). La phosphocréatine se lie à l'adénosine diphosphate pour le reconvertir en ATP (adénosine triphosphate), une importante source d'énergie cellulaire.
La créatine provoque une rétention intra-musculaire. De par ses propriétés stimulantes, elle est utilisée dans le sport de compétition comme produit dopant (par exemple en haltérophilie) car elle améliore la performance des muscles en effort anaérobie. La prise de créatine augmente le niveau de phosphocréatine dans les muscles jusqu'à 20%, rendant ceux-ci plus efficaces en effort intense et rapide.
Les revendeurs de créatine tendent à soutenir que la créatine augmente la masse musculaire, or il s'agit plus d'une augmentation de la masse d'eau dans le muscle transportée par les molécules de créatine. De ce fait, la prise de créatine entraîne un risque de déshydratation. Prendre de la créatine sur une courte période est sans danger, mais on ne connait toujours pas les effets d'une prise sur le long terme.
L'usage et la vente de créatine est illégal en France, non par décret mais parce qu'elle n'a pas d'existence légale, et aucune autorisation d'emploi n'a été accordée. Cependant sa vente et son usage sont légaux dans la plupart des autres pays. Cela peut poser des problèmes d'équité lors de compétitions internationales.
